The 'Spectrum' singer has been advised to rest by doctors on the eve of this year's festival.
Florence & The Machine have been forced to cancel their appearance at this week's Benicassim festival due to illness.
In a statement, organisers said singer Florence Welch was suffering from "acute vocal strain causing swelling of the vocal cords".
The singer was struck down with the problem after a recent performance in Ireland and has been advised to rest by doctors.
Organisers said: "Given the sudden and unexpected illness suffered by Miss Welch, FIB 2012 organization has been unable to find a substitute in such a small time frame."
This year's Benicassim is due to take place from 12th July – 15th July in Spain.
Meanwhile, Florence's new single 'Spectrum', which has been remixed by Calvin Harris, is on course to top the Vodafone Big Top 40 this weekend.
