The boy, who has not yet been named, died in hospital on Monday, Staffordshire Police said.
His mother and two more of her children, a girl aged four and an 18 month old boy, were also taken to hospital following the crash on Sunday but were later discharged with minor injuries.
The crash happened on the southbound carriageway of the M6 between the two slip roads for Hilton Services near Cannock and involved a grey Seat Leon and a silver Mercedes C- Class.
